the gearbox bolt pattern is different, if thats what you mean? all I4s are one bolt pattern, all v6s are a different one.
to keep your current prop, etc. youll need to find a mt75 for a v6 application (which are rare, I'm pretty sure they were never a uk option or something), or make a hybrid box using a xr4x4 box with the transfer box replaced by 2wd bits- i know its do-able, but I'm not entirely sure how as ive never done it.
otherwise, youre looking at going type9 and having to change the prop and gearbox mount too.
the rest is all pretty straightforward.

Transit mt75 is different to the car one. There's a adopter plate to bolt the engine to the box which I'm hoping to mod to fit the v6

It's mainly ambulances that were 2.9, keep an eye out for one it will have all you need, like engine mounts and adapter but as you'll need a V6 box you might find it easyer nd cheaper to buy a compleat amby ? Ive heard of peeps moding the pinto plate for a V6 but you still need the V6 box

Transit gearbox has the gear stick coming out the side/top, a car box will put the stick and 18" to far back i would say, theres loads of room in a transit engine bay, mounts can be fabbed pretty easy, auto gearbox would make life much easier, but the 2.9 is only 150bhp, in a transit with an auto on it it wont feel fast.
If your happy with an auto box then a 24v is a better bet, will still fit the length of the engine bay better than a V8 and has 200bhp so will feel brisk even with the stock auto on it. I'd be looking at a higher geared axle as well.

Why can't you just change the bell housing over, wouldn't that make things a lot easier? I don't know if you can, but it's a thought as I have never done this either, but surely the bell housing just bolt on using 4(?) bolts.
Cheers,
Stu.

mt75 boxes don't have removeable bellhousings, its part of the gearbox front casing the 24v will fit, but needs the bulkhead 'adjusting' to clear the monstrous alloy heads when fitted in the transit you may be lucky and find the gearbox in the transit has an adapter bolted to it already, as some of the diesel engined ones were like that. if thats the case, just redrill the plate to bolt to the cologne engine

The LT77, or the R380 spring to mind Stu I don't think either were ever fitted to the ford V6 though? Shame, as they're quite nice 'boxes
